| Abstract | Malignant mesothelioma, a tumor of the serosal lining of the pleura, pericardium and peritoneum, is a worldwide problem. Simian virus 40 (SV40) has been implicated as a potential co-factor in the etiology of this disease, but despite many studies, its presence is still controversial. To address this controversy, we studied SV40 DNA presence in 52 pleural malignant mesotheliomas using real time PCR (Q-PCR) and Digital PCR. Digital PCR is a new method developed to address the potential problem of low SV40 copy numbers in tumors by improving SV40 DNA. The SV40 positivity rate with these assays was 25-35%, indicating that SV40 is present in some malignant mesotheliomas. Additionally, our Digital PCR results indicate that this new method is useful to study SV40 presence in different human tumors as it is more sensitive, allowing detection of low SV40 copy numbers. |
