Abstract
Anxiety disorders are a series of chronic mental diseases that affect many people all over the world, which are mainly characterized by significant and uncontrollable feelings of anxiety and depressed. During the COVID-19, people’s lives have been greatly affected. Long-term quarantine at home and lack of social activities have led to getting anxiety disorders more easily. Dihydromyricetin (DHM), a kind of bioactive flavonoid, shows the effects on decreasing alcohol usage, reducing anxiety-related behaviors and repairing cognition impairment in our team's previous studies. In current study, using a social isolation mice model established in our lab, we found that isolated animals showed increased anxiety levels in elevated plus maze and open field behavior assays. We also found a decreased brain-derived neurotrophic factor (BDNF) level as well as activated astrocytes, glial cells which stabilize synaptic connection and maintain synaptic function. While DHM can restore the BDNF concentration to a normal level and also alleviates the activation of astrocyte by decreasing the number of astrocyte and reducing the branch number. Our study suggested that short term of social isolation could induces astrocyte overactivation and BDNF reduction, both contribute to synaptic functions. DHM can restore the synapse dysfunction and reduce anxiety level, suggesting DHM can be considered as a novel candidate for treating anxiety disorders.
