CHAPTER I
INTRODUCTION
The p r im a r y p u r p o s e o f t h i s i n v e s t i g a t i o n was t o
d e te r m in e th e c a u s e o f t h e d i f f e r e n c e i n b e h a v i o r b e tw e e n
serum c h o l i n e s t e r a s e i n l i v e r damage i n d i s e a s e d hum ans
and i n r a b b i t s w i t h e x p e r i m e n t a l l y c a u s e d l i v e r dam age.
A s e c o n d a r y o b j e c t i v e w as t h e f u r t h e r e l u c i d a t i o n o f t h e
n a t u r e o f r a b b i t c h o l i n e s t e r a s e . Num erous o b s e r v e r s hav e
n o te d t h a t i n human l i v e r dam age, p a r t i c u l a r l y c i r r h o s i s ,
serum c h o l i n e s t e r a s e v a l u e s a r e d e p r e s s e d b e lo w n o rm a l
l e v e l s . R e c e n t w ork on v a r i o u s s p e c i e s o f a n im a ls i n
w h ic h l i v e r damage h a s b e e n p ro d u c e d b y p o i s o n i n g w i t h
c a rb o n t e t r a c h l o r i d e h a s n o t p ro d u c e d c o r r e s p o n d i n g r e
s u l t s .
I t a p p e a re d t h a t t h e r e m ig h t be two p o s s i b l e
e x p l a n a t i o n s f o r t h e s e a p p a r e n t l y c o n t r a d i c t o r y phenom ena;
f i r s t , t h a t t h e e x p e r i m e n t a l a n im a ls w e re n o t b r o u g h t t o
a c i r r h o t i c s t a t e s i n c e t h e r e p o r t e d w ork w as o f s h o r t
d u r a t i o n , and s e c o n d , t h a t t h e enzyme s y s te m s o f t h e
s p e c i e s m ig h t be d i f f e r e n t .
The e x p e r i m e n t a l p a r t o f t h i s w o rk w as p la n n e d
a c c o r d i n g l y . R a b b i t s w e re f e d c a rb o n t e t r a c h l o r i d e and
t h e c o u r s e o f l i v e r damage w as fo llo w e d b y serum c h o l i n
e s t e r a s e d e t e r m i n a t i o n s and h i s t o l o g i c a l e x a m in a tio n —

was com pared t o n o rm al human seru m w i t h r e g a r d t o s u b s t r a t e
s p e c i f i c i t y a n d b e h a v i o r i n th e p r e s e n c e o f i n h i b i t o r s .

CHAPTER II
HISTORICAL REVIEW
The p i o n e e r w ork o f D a le on a c e t y l c h o l i n e i n 1914
s u g g e s te d t h a t a n enzym e was p r o b a b ly in v o lv e d i n c o n t r o l l
in g th e am ount o f t h i s s u b s ta n c e i n t h e c i r c u l a t i o n . I n
1926 Loew i fo u n d t h a t a c e t y l c h o l i n e a s w e l l a s t h e s u b s ta n c e
l i b e r a t e d on v a g u s s t i m u l a t i o n w ere b o t h i n a c t i v a t e d by
i n c u b a t i o n w ith h e a r t e x t r a c t . H e a te d o r i r r a d i a t e d h e a r t
e x t r a c t h a d no su c h e f f e c t . F u r t h e r w ork by L oew i and
N a v r a t i l on t h e e f f e c t o f p h y s o a tig m in e on h e a r t l e d t o
t h e c o n c l u s i o n t h a t t h e e f f e c t o f t i l l s a l k a l o i d was t h e
i n h i b i t i o n o f t h e a g e n t w hich d e s t r o y e d a c e t y l c h o l i n e ,

I . PROPERTIES OF THE CHOLINESTERASES
The e a r l y c h a r a c t e r i z a t i o n o f t h e enzyme s y s te m s
in v o lv e d i n c h o li n e m e ta b o lis m may be f o u n d i n t h e w ork o f
S ted m an a n d c o - w o r k e r s . They s t u d i e d th e i n h i b i t o r y a c t i o n
o f c e r t a i n p h y s o a tig m in e a n a lo g u e s on l i v e r e s t e r a s e and

t i t r a t i o n m ethod i n a s t u d y o f t h e h y d r o l y s i s o f th e t h r e e
e s t e r s b y t h e s e r a o f v a r i o u s s p e c i e s . They fo u n d a c e t y l

I n 1935 Stedm an an d S tedm an d e s c r i b e d a m an o m etric
d e t e r m i n a t i o n o f c h o l i n e s t e r a s e u s i n g a B a n c r o f t a p p a r a t u s ,
an d d e m o n s tr a te d a c e t y l c h o l i n e s t e r a s e i n r e d b lo o d c e l l s
( 3 3 ) . E a s s o n and Stedm an (1 1 ) b y m eans o f i n h i b i t i o n and
m ixed s u b s t r a t e s t u d i e s f u r t h e r e d t h e d i f f e r e n t i a t i o n o f
" e s t e r a s e " i n t o c h o l i n e s t e r a s e a n d m e th y l b u t y r a s e , and
fo u n d o n ly th e f o r m e r i n human se ru m .
A l l e s a n d Hawes (2 ) i n t r o d u c e d t h e g l a s s e l e c t r o d e
c o n ti n u o u s t i t r a t i o n m ethod f o r c h o l i n e s t e r a s e d e t e r m i n a t i o n
a n d d e m o n s tr a te d a n o t a b l e q u a l i t a t i v e d i f f e r e n c e b e tw e e n
t h e c h o l i n e s t e r a s e s o f r e d c e l l s and se ru m . The enzym es
h e h a v e d d i f f e r e n t l y w i t h r e g a r d to a c e t y l c h o l i n e d e r i v a
t i v e s , s u b s t r a t e c o n c e n t r a t i o n s , pH a c t i v i t y c u r v e s , a n d
s a l t c o n c e n t r a t i o n .
M endel and Hudney (2 4 ) d e m o n s tr a te d t h e p r e s e n c e
o f two d i f f e r e n t c h o l i n e s t e r a s e s w h ic h th e y d e s i g n a t e d a s
" t r u e " an d "pseudo" c h o l i n e s t e r a s e s , th e f o r m e r b e in g
s p e c i f i c f o r c h o li n e e s t e r s and th e l a t t e r b e in g a b le t o
s p l i t c h o l i n e e s t e r s a s w e l l a s o t h e r e s t e r s . I t was
d e m o n s tr a te d t h a t A lle s * c e l l and serum c h o l i n e s t e r a s e s
w e re r e s p e c t i v e l y t r u e c h o l i n e s t e r a s e an d p se u d o c h o l i n —
e s t e r a s e . I t w as fo u n d t h a t a t h ig h s u b s t r a t e c o n c e n t r a t i o n s
th e a c t i o n o f th e p s e u d o - c h o l i n e s t e r a s e p re d o m in a te d w h ile
th e t r u e c h o l i n e s t e r a s e w as m ore a c t i v e a t low a c e t y l c h o l i n e
c o n c e n t r a t i o n s .
A more c o n c r e t e d e m o n s tr a ti o n o f the p r e s e n c e o f two
enzym es c a p a b le o f h y d r o l y z i n g a c e t y l c h o l i n e was made b y
M e n d el, M u n d e ll, and R udney ( 2 3 ) . I t was fo u n d t h a t t h e
t r u e c h o l i n e s t e r a s e h y d r o ly z e d a c e t y l ■ ^ g -m e th y lc h o lin e
w h ile p s e u d o - c h o l i n e s t e r a s e h a d n o e f f e c t on i t , a n d t h a t
t h e p s e u d o - c h o l i n e s t e r a s e a lo n e c o u ld h y d r o ly z e b e n z o y l
c h o l i n e . F o r t u n a t e l y , n e i t h e r of t h e s e e s t e r s w ere a t t a c k e d

toy t h e common e s t e r a s e . I t w as a l s o n o t e d t h a t t h e d i s t r i
b u t i o n o f t h e c h o l i n e s t e r a s e d i f f e r e d i n t h e p lasm a o f
v a r i o u s s p e d i e s . The t r u e c h o l i n e s t e r a s e was p r e s e n t i n
p la sm a o f . a l l s p e c ie s * w h ile t h e p s e u d o - c h o l i n e s t e r a s e v a r i e d
fro m a c o m p le te a b s e n c e t o p re d o m in a n c e i n t h e v a r i o u s
s p e c i e s i n v e s t i g a t e d .
The s i t u a t i o n was f u r t h e r c o m p lic a te d toy Saw yer (>30)
who c o n c lu d e d t h a t t h e r e w a s, i n th e l i v e r s o f t h e g u in e a
p i g and r a b b i t , a n enzyme c a p a b le o f h y d r o l y z i n g b e n z o y l
c h o lin e b u t n o t a c e t y l c h o l i n e . T h is he te r m e d ’’b e n z o y l
c h o l i n e s t e r a s e . ”
H aw kins an d G u n te r (1 9 ) d e t a i l e d a m ethod u s i n g t h e
i n h i b i t o r e s e r i n e t o d e te r m in e w h e th e r th e y w e re d e a l i n g
w ith p s e u d o - c h o l i n e s t e r a s e o r b e n z o y l c h o l l n e s t e r a s e .
F u r t h e r d i f f e r e n t i a t i o n o f t h e c h o l i n e s t e r a s e s was
made b y H aw kins and M en d el, (2 0 ) u s in g th e i n h i b i t o r
d i l s o p r o p y l f l u o r o p h o s p h a t e . DFP a t low c o n c e n t r a t i o n s
w as fou nd t o s p e c i f i c a l l y i n h i b i t t h e p s e u d o - c h o l i n e s t e r a s e .
I I . HISTOLOGY OF LIVER POISONED BY CARBON TETRACHLORIDE.
The c l a s s i c w ork i n t h i s f i e l d was t h e s tu d y made b y
Cam erdn i n 1936 (9)>, v«ho f o llo w e d t h e c o u r s e o f l i v e r damage
i n t h e r a t . I n t h i s p a p e r t h e w ork o f some e a r l y i n v e s t i g a t o r s
w ith r e g a r d to t h e r a b b i t was re v ie w e d ( 1 9 , 2 8 , 3 8 ) , w h ic h
f i t i n w e l l w ith C am ero n ’ s f i n d i n g s . The s t u d i e s by s e v e r a l

o t h e r w o r k e rs i n v a r i o u s s p e c i e s w ere a l s o i n g e n e r a l a g r e e -
tnent w i t h Cam eron ( 7 , 1 0 , 1 4 , 1 5 , 1 6 , 3 7 ) . V a r i a t i o n s among
d i f f e r e n t s p e c i e s a r e e n c o u n te r e d w ith r e g a r d to am ount o f
l e t h a l d o s e , t o l e r a n c e , tim e f o r n e c r o s i s to a p p e a r , an d
l e n g t h o f r e c o v e r y p e r i o d . The h i s t o l o g i c a l p i c t u r e , a s
i n d i c a t e d a b o v e , i s f a i r l y c o n s t a n t . T h e r e f o r e , a s u r v e y
o f C a m e ro n 's w ork w i l l s e r v e t o o u t l i n e t h e t y p i c a l h i s t o
l o g i c a l c h a n g e s i n a l i v e r dam aged b y c a rb o n t e t r a c h l o r i d e .
One d a y a f t e r a s i n g l e d o se o f c a rb o n t e t r a c h l o r i d e ,
0 .0 1 6 - 0 .0 3 3 gram s p e r k i l o o f body w e i g h t , t h e l o b u l a r
p a t t e r n o f th e l i v e r a p p e a re d e x a g g e r a t e d . The c e l l s n e a r
t h e c e n t r a l v e i n s t a i n e d h o m o g e n e o u sly , b u t w ere d i f f e r e n t
f r o m t h e p e r i p h e r a l c e l l s and o f t e n showed p y k n o t ic o r
d e g e n e r a t e d n u c l e i . T hese c e l l s w e re b o r d e r e d b y e n l a r g e d
t h r e a d l i k e s t r u c t u r e s . N um erous m it o s e s w ere o b s e rv e d #
T h ere w as c o n g e s t i o n o f c e n t r a l a n d m id - z o n a l a r e a s , h e m o rr
h a g e , and o e l l u l a r i n f i l t r a t i o n among t h e n e c r o t i c c e l l s b y
ly m p h o c y te s an d h i s t o c y t e s .
T h ree d a y s a f t e r th e i n j e c t i o n , t h e l o b u l a r c e l l s
w ere g o n e , l e a v i n g o n ly m esenchym al s tro m a . T h e re w as some
ly m p h o c y tic i n f i l t r a t i o n . The c e l l s o f t h e m id - z o n a l r e g i o n
showed f a t t y i n f i l t r a t i o n . No g ly c o g e n was o b s e r v e d . T h ere
was a l a r g e num ber of m i t o s e s . I n t h e p o r t a l t r a c t s t h e r e
w ere ly m p h o c y te s , young f i b r o b l a s t s , an d some p o ly m o rp h s .

2 . RESULTS
S t a b i l i t y o f th e enzym e. The s t a b i l i t y o f t h e r e
f r i g e r a t e d enzyme h a s b e e n n o te d b y s e v e r a l w o rk e rs ( 2 , 6 ,
3 3 ) , and was f u r t h e r t e s t e d i n t h i s w o rk . A sp e cim en o f
serum was s t o r e d i n t h e r e f r i g e r a t o r o v e r a p e r i o d o f two
w eeks and t e s t e d a t i n t e r v a l s . No s i g n i f i c a n t c h a n g e s w ere
o b s e r v e d .

p o i s o n i n g , f o llo w e d b y a s lo w , i r r e g u l a r r e t u r n t o n o r m a l.
