| Abstract | Background: Cleft lip and palate are common human birth defects which arise from many etiologies, including the following: genetic disorders, chromosomal abnormalities, teratogenic exposures, sporadic, and unknown causes. The causes are being tested through animal model studies. However, no previous review has specifically compared the morphology of mouse cleft palates with the same defect in humans.; Purpose: To compare the genetic causes similar to mice and humans. To test the hypothesis that the specific causes of mouse cleft are also identified in humans.; Summary: Mutations in TGFβ3, MSX1, IRF6, and P63 all cause cleft lip and or palate in humans. Only single sporadic human cases of mutations in TGFβ-IR, TGFβ-IIR, and PAX9 were found to be associated with clefting. This review validates the mouse model and contributes to a greater understanding of human craniofacial anomalies. |
