Abstract | Galectins are lectins which posscess carbohydrate recognition domains that bind ß-galactosides. Secreted alkaline phosphatase (SEAP) is a secreted form of alkaline phosphatase, commonly used as surrogate marker for tumor burden measurement. When expressing SEAP, galectin-3 is down-regulated in ovarian carcinoma cell line ovcar3. Since galectin-3 is known to confer survival advantages to cancer cells, it would be worthwhile to confirm the effect on different cell lines and to elucidate how SEAP causes the down-regulation of galectin-3. This thesis focuses on the construction of SEAPexpressing mammalian expression vectors that could be used as tools to investigate the mechanisms of galectin-3 regulation by SEAP. Two vectors were constructed, one based on a constitutive promoter and the other on an inducible promoter. Also, the ovarian carcinoma cell line HEY was prepared for use in the inducible system by infection with the ecdysone receptor proteins, and was tested for effective induction. |
